Western blot was performed on whole cell (25 \u00b5g, lane 1) and histone extracts (15 \u00b5g, lane 2) from HeLa cells, and on 1 \u00b5g of recombinant histone H2A, H2B, H3 and H4 (lane 3, 4, 5 and 6, respectively) using the Bioss antibody against H3K9me3 (Cat. No. bs-53114R). The antibody was diluted 1:1,000 in TBS-Tween containing 5% skimmed milk.
HeLa cells were stained with the Bioss antibody against H3K9me3 (bs-53114R) and with DAPI. Cells were fixed with 4% formaldehyde for 10\u2019 and blocked with PBS\/TX-100 containing 5% normal goat serum and 1% BSA. The cells were immunofluorescently labeled with the H3K9me3 antibody (middle) diluted 1:1,000 in blocking solution followed by an anti-rabbit antibody conjugated to Alexa488. The left panel shows staining of the nuclei with DAPI. A merge of both stainings is shown on the right.
To determine the titer of the antibody, an ELISA was performed using a serial dilution of the antibody directed against human H3K9me3 (bs-53114R) in antigen-coated wells. The antigen used was a peptide containing the histone modification of interest. By plotting the absorbance against the antibody dilution, the titer of the antibody was estimated to be 1:30,000.
A Dot Blot analysis was performed to test the cross-reactivity of the antibody against H3K9me3 (Cat. No. bs-53114R) with peptides containing other modifications and unmodified sequences of histone H3 and H4. One hundred to 0.2 pmol of the peptide containing the respective histone modification were spotted on a membrane. The antibody was used at a dilution of 1:20,000. The figure shows a high specificity of the antibody for the modification of interest.

Dot Blot analysis was performed with peptides containing a variety of histone modifications and the unmodified H4. 0.2-100 pmol of the respective peptides were spotted on a membrane. H4K5ac Polyclonal Antibody (bs-53099R) was used at 1:5,000 dilution. Result shows high specificity of the antibody for the modification of interest.
ChIP were performed using HeLa cells using H4K5ac Polyclonal Antibody (bs-53099R). Chromatin was sheared from 1 million cells. A titration of the antibody consisting of 1, 2, 5, and 10\u03bcg per ChIP experiment was analyzed. IgG (2\u03bcg\/IP) was used as negative IP control. Quantitative PCR was performed with primers for promoter of the active gene EIF4A2 and for a region 1 kb upstream of the GAPDH as positive controls, and for the inactive MYOD1 gene and Sat2 satellite repeat region used as negative controls. Result shows the recovery, expressed as a % of input (the relative amount of immunoprecipitated DNA compared to input DNA after qPCR analysis).

Western blot analysis was performed on 100 fmol ER alpha (ERa) and ER beta (ERb1) recombinant protein with ER alpha (5G7) Monoclonal Antibody (bsm-53007M) at a concentration of 7 \u03bcg\/ ml. Figure shows that the antibody is specific for ER alpha isoform, whereas the ER beta isoform is not recognized.
COS-7 cells transiently over-expressing human ER\u03b1 (left) or ER\u00df1 (right) were labeled with the ER alpha (5G7) Monoclonal Antibody (bsm-53007M), used at a concentration of 15 \u03bcg\/ml, followed by a biotinylated secondary antibody and peroxidase-labeled avidin. Figure shows the specificity of the antibody for the ER alpha isoform.
ChIP assays was performed using MCF7 cells treated with the ER agonist estradiol for 3 hours prior to harvesting, ER alpha (5G7) Monoclonal Antibody (bsm-53007M), and optimized primer sets for qPCR. Sheared chromatin from 3 million cells and 5 \u03bcg of antibody were used per ChIP experiment. Recovery (%: ChIP\/input) and occupancy (x fold: +ve\/-ve) are shown. QPCR was performed with primers for the GREB1 promoter and for exon 2 of the myoglobin gene used as a negative control. Figure shows the recovery (the relative amount of immunoprecipitated DNA compared to input DNA) and the occupancy (ratio +\/- control target). These results demonstrate the occupancy of the GREB1 promoter by ERalpha.

The membrane was probed with the primary antibody at dilutions of 1:100 (lane 1), 1:250 (lane 2), 1: 500 (Lane 3) and 1:1,000 (Lane 4). The identity of the bands at ~95 kDa is unknown but may be degraded TAF1.
