Mouse Fatty Acid Transport Protein 4 (FATP4) ELISA Kit
Principle of the Assay
The microtiter plate provided in this kit has been pre-coated with an antibody specific to FATP4. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to FATP4. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After the TMB substrate solution is added, only those wells that contain FATP4, biotin-conjugated antibody, and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution, and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of FATP4 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Target Information
Mediates the import of long-chain fatty acids (LCFA) into the cell by facilitating their transport across cell membranes (PubMed:10518211, PubMed:20448275). Appears to be the principal fatty acid transporter in small intestinal enterocytes (PubMed:20448275). Also functions as an acyl-CoA ligase catalyzing the ATP-dependent formation of fatty acyl-CoA using LCFA and very-long-chain fatty acids (VLCFA) as substrates, which prevents fatty acid efflux from cells and might drive more fatty acid uptake (By similarity). Plays a role in the formation of the epidermal barrier. Required for fat absorption in early embryogenesis (PubMed:11404000, PubMed:12821645, PubMed:15653672, PubMed:15699031). Probably involved in fatty acid transport across the blood barrier (By similarity). Indirectly inhibits RPE65 via substrate competition and via production of VLCFA derivatives like lignoceroyl-CoA. Prevents light-induced degeneration of rods and cones (PubMed:23407971).

Performance Characteristics
REPEATABILITY |
Intra-assay Precision (Precision within an assay): 3 samples with low, middle, and high-level FATP4 were tested 20 times on one plate, respectively. |
SENSITIVITY | The minimum detectable dose was 0.078ng/mL. |
ASSAY RANGE | 0.156-10ng/mL |
SPECIFICITY | This assay has high sensitivity and excellent specificity for the detection of FATP4. No significant cross-reactivity or interference between FATP4 and analogs was observed. Note: Limited by current skills and knowledge, it is impossible to perform all possible cross-reactivity detection tests between FATP4 and all analogs, therefore, cross reactivity may still exist. |
