Rat Apelin (APLN) ELISA Kit

Principle of the Assay

The microtiter plate provided in this kit has been pre-coated with an antibody specific to APLN. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to APLN. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After the TMB substrate solution is added, only those wells that contain APLN, biotin-conjugated antibody, and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution, and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of APLN in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Target Information

Endogenous ligand for the apelin receptor (APLNR) (PubMed:11336787, PubMed:11359874, PubMed:26611206). Drives internalization of the apelin receptor (PubMed:11359874). Apelin-36 dissociates more hardly than (pyroglu)apelin-13 from APLNR (PubMed:11336787). Hormone involved in the regulation of cardiac precursor cell movements during gastrulation and heart morphogenesis (By similarity). Has an inhibitory effect on cytokine production in response to T-cell receptor/CD3 cross-linking; the oral intake of apelin in the colostrum and the milk might therefore modulate immune responses in neonates (By similarity). Plays a role in early coronary blood vessels formation (By similarity). Mediates myocardial contractility in an ERK1/2-dependent manner (PubMed:26611206). May also have a role in the central control of body fluid homeostasis by influencing vasopressin release and drinking behavior (PubMed:10617103, PubMed:11359874).

Storage

The TMB Substrate, Wash Buffer (30X concentrate), and the Stop Solution should be stored at 4°C upon receipt, while the other items should be stored at -20°C.

Performance Characteristics

REPEATABILITY	Intra-assay Precision (Precision within an assay): 3 samples with low, middle, and high-level APLN were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle, and high-level APLN were tested on 3 different plates, with 8 replicates in each plate. SENSITIVITY	The minimum detectable dose was 5.62pg/mL.
ASSAY RANGE	15.6-1000pg/mL
SPECIFICITY	This assay has high sensitivity and excellent specificity for the detection of APLN. No significant cross-reactivity or interference between APLN and analogs was observed. Note: Limited by current skills and knowledge, it is impossible to perform all possible cross-reactivity detection tests between APLN and all analogs, therefore, cross reactivity may still exist.
