L1 mouse kidney lysates L2 mouse brain lysates probed with Anti ER-beta Polyclonal Antibody, Unconjugated (bs-0116R) at 1:200 overnight at 4˚C. Followed by conjugation to secondary antibody (bs-0295G-HRP) at 1:3000. Predicted band 58kD. Observed band size: 58kD

HeLa cells probed with Estrogen receptor beta Polyclonal Antibody, Unconjugated (bs-0116R) at 1:100 for 30 minutes followed by incubation with a conjugated secondary (bs-0295G-PE) (green) for 30 minutes compared to control cells (blue), secondary only (light blue) and isotype control (orange).

Lane 1: Mouse Testis lysates; Lane 2: Rat Testis lysates; Lane 3: Mouse Ovary lysates; Lane 4: Mouse Uterus lysates ; Lane 5: Rat Uterus lysates probed with Estrogen receptor beta Polyclonal Antibody, Unconjugated (bs-0116R) at 1:1000 dilution and 4˚C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.

$A431 cells were fixed with 4% PFA for 10min at room temperature,permeabilized with 90% ice-cold methanol for 20 min at -20\u2103, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with \r\nEstrogen receptor beta Antibody(bs-0116R)at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).$

Background	Nuclear hormone receptor. Binds estrogens with an affinity similar to that of ESR1, and activates expression of reporter genes containing estrogen response elements (ERE) in an estrogen-dependent manner. Isoform beta-cx lacks ligand binding ability and has no or only very low ere binding activity resulting in the loss of ligand-dependent transactivation ability. DNA-binding by ESR1 and ESR2 is rapidly lost at 37 degrees Celsius in the absence of ligand while in the presence of 17 beta-estradiol and 4-hydroxy-tamoxifen loss in DNA-binding at elevated temperature is more gradual.
