$THP-1 (H) treated with PMA (100 ng\/mL) for 24 h, Replace with fresh complete culture medium and continue to culture for 24 h, + hIFN-\u03b1 (500U\/mL) for 24 h, then with Brefeldin A (1\u00b5g\/ml) for 3 h, 25 \u03bcg total protein per lane of cell lysates (see on figure) probed with CHI3L1 monoclonal antibody, unconjugated (bsm-55783R) at 1:1000 dilution and 4\u00b0C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.$
