Total Thiol Content Assay Kit

Overview

This kit measures total thiol content rather than the activity of antioxidant enzymes. The color-forming reaction with DTNB detects thiol groups collectively, including those associated with GSH and proteins, rather than identifying each thiol-containing compound individually.

Thiol groups, including reduced glutathione and protein thiols, play central roles in redox signaling, reactive oxygen species scavenging, and repair of oxidatively damaged proteins. Total thiol content can be used with a separate GSH measurement to estimate protein thiols.

Assay principle

Thiol groups react with 5,5′-dithiobis(2-nitrobenzoic acid) to produce a yellow compound with a maximum absorbance at 412 nm.
