IL-4R Polyclonal Antibody

Formalin-fixed and paraffin embedded human colon carcinoma labeled Anti-IL-4R\/CD124 Polyclonal Antibody, Unconjugated (bs-2458R) at 1:200 followed by conjugation to the secondary antibody Goat Anti-Rabbit IgG, AF488 conjugated(bs-0295G-AF488)used at 1:200 dilution

Tissue\/cell: A431 cells; 4% Paraformaldehyde-fixed; Triton X-100 at room temperature for 20 min; Blocking buffer (normal goat serum) at 37\u00b0C for 20 min; Antibody incubation with IL4R Polyclonal Antibody, Unconjugated (bs-2458R) at 1:100 for 90 minutes at 37\u00b0C; followed by a conjugated Goat Anti-Rabbit IgG antibody with FITC conjugation (bs-0295G-FITC) at 37\u00b0C for 90 minutes, DAPI (5ug\/ml, blue) was used to stain the cell nuclei.

Human Raji Cells were fixed with 2% PFA (10min at room temperature). The cells were then incubated in 1 X PBS containing 0.5% BSA + 10% goat serum to block non-specific protein-protein interactions for 15 min at room temperature. Staining with IL4R Polyclonal Antibody, Unconjugated (bs-2458R) was performed at 1:100 for 30 min at room temperature. The secondary antibody, Goat anti-rabbit IgG-PE, was used at a 1:200 dilution for 40 min at room temperature. Primary antibody staining (green) is compared to compared to unstained cells (dark blue), secondary only (light blue), and isotype control (orange).

25 ug total protein per lane of various lysates (see on figure) probed with IL-4R polyclonal antibody, unconjugated (bs-2458R) at 1:1000 dilution and 4\u00b0C overnight incubation. Followed by conjugated secondary antibody incubation at r.t. for 60 min.
