$ChIP assays were performed using the TIP5 antibody (bs-53052R). Chromatin from HEK293T and NIH3T3 cells were formaldehyde cross-linked and sheared to yield fragments with an average length of 200 to 400 bp. ChIP was performed overnight at 4\u00b0C with 100 \u03bcg sheared chromatin and either 5 \u03bcl of the TIP5 antibody or 5 \u03bcl IgG which was used as negative IP control. The IP\u2019d DNA was analyzed by qPCR with primer sets for the promoter and the coding region of the 28s ribosomal RNA gene. The figure shows the recovery by the TIP5 antibody and by IgG (set to 1), normalized to the input DNA. These results show that, both in HEK293T and in NIH3T3 cells, TIP5 is associated with the promoter, but not with the coding region of the 28srRNA gene.$

Background	Essential component of the NoRC (nucleolar remodeling complex) complex, a complex that mediates silencing of a fraction of rDNA by recruiting histone-modifying enzymes and DNA methyltransferases, leading to heterochromatin formation and transcriptional silencing. In the complex, it plays a central role by being recruited to rDNA and by targeting chromatin modifying enzymes such as HDAC1, leading to repress RNA polymerase I transcription. Recruited to rDNA via its interaction with TTF1 and its ability to recognize and bind histone H4 acetylated on 'Lys-16' (H4K16ac), leading to deacetylation of H4K5ac, H4K8ac, H4K12ac but not H4K16ac. Specifically binds pRNAs, 150-250 nucleotide RNAs that are complementary in sequence to the rDNA promoter; pRNA-binding is required for heterochromatin formation and rDNA silencing.
