SH-SY5Y cells were fixed with 4% PFA for 10min at room temperature,permeabilized with 90% ice-cold methanol for 20 min at -20\u2103, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with Thioredoxin Polyclonal Antibody(bs-0458R)at 1:100 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature. Human U937 Cells were fixed with 2% PFA (10min at room temperature). The cells were then incubated in 1X PBS containing 0.5% BSA + 10% goat serum to block non-specific protein-protein interactions for 15 min at room temperature. Staining was performed with CD86 Polyclonal Antibody, Unconjugated (bs-1035R) at 1:100 for 30 min on ice. A Goat Anti-rabbit IgG-PE secondary antibody was used for a 30 min incubation on ice. Primary antibody staining (green) is compared to unstained cells (dark blue), secondary antibody only (light blue) and isotype control (bs-0295P; orange).

Formalin-fixed and paraffin embedded mouse lymphoma tissue labeled Anti-CD3 Polyclonal Antibody, Unconjugated (bs-0765R) 1:200, followed by conjugation to the secondary antibody Goat Anti-Rabbit IgG, FITC conjugated(bs-0295G-FITC)used at 1:200 dilution for 40 minutes at 37\u00b0C.
Jurkat (Positive) and A431 (Negative control) cells (black) were incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with CD3 epsilon Polyclonal Antibody (bs-0765R) at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2% BSA in PBS, followed by secondary antibody(blue) incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).
Lane 1: Recombinant mouse CD3E protein, C-mFc (HEK293)(bs-43509P) probed with CD3E Polyclonal Antibody, Unconjugated (bs-0765R) at 1:1000 dilution and 4\u00b0C overnight incubation. Followed by conjugated secondary antibody incubation at 1:20000 for 60 min at 37˚C.
